Anyone seen this red plug before? I don't recognize it but I don't have another 5-spd around right now to compare it to.

Looks like someone used some junk box components to replace the plug. Most likely not a metric thread. On my plug I welded a nut on the OEM plug and left it that way.

It means that the shaft nut backed off and drilled a hole in the rear cover, and someone plugged the hole. More usual repair is to weld it up.
There is not supposed to be a plug or hole there at all.
ON closer look that hole is kind of in the wrong place for the shaft nut problem, so "never mind". Dave M is correct.

So that plug is just a weak-sauce replacement for a bolt?
Dave - is the hole drilled by the backed off nut more on the butt end?

Here's a shot from the inside. I didn't have a photo of a welded cover handy. But you can see where the nut started drilling the housing.

I don't have a manual trans handy either but if you look at 4-1-1 in the parts manual there is no plug there. My guess is someone at some time removed the control shaft and removed the blanking plug that was pressed into the housing. To close it up they used that ugly looking method. If it isn't leaking you can leave it alone. If you want to make it pretty you could remove it and try to find something better.

And for even more reference, here's a photo of one (mine) where the nut made it through the housing and the housing was weld repaired:
